MicroRNA-138 suppresses cell proliferation, migration and invasion by targeting smoothened (SMO) in hepatocellular carcinoma

چکیده
Hedgehog (Hh) signaling pathway is a crucial signal transduction pathway that associated with tumorigenesis in multiple human tissues. Recently, microRNA-138 (miR-138) has been reported to be frequently involved in different tumorigenesis, including hepatocellular carcinoma (HCC). However, whether miR-138 was correlated with Hedgehog (Hh) signaling pathway implicated in the progression of HCC remains unclear. Here, we demonstrated that the expression of miR-138 was downregulated in HCC tissues compared with their matched adjacent nontumor tissues (p < 0.001). Overexpression of miR-138 leaded to impaired cell viability, colony formation, cell migration and invasion of HepG2 cells. Furthermore, smoothened (SMO) was identified as a novel target of miR-138, and the expression of SMO was inhibited by miR-138 overexpression in HepG2 cells. Besides, SMO was upregulated in HCC tissues compared to the matched adjacent normal tissues and its downregulation photocopied the effects of miR-138 overexpression in HepG2 cells. These findings indicate that miR-138 functioned as a tumor suppressor partially via repressing SMO expression in HCC.

Objective(s): MicroRNAs (miRNAs) are considered as powerful, post-transcriptional regulators of gene expression in hepatocellular carcinoma cells (HCC). However, the function of miR-92a is still unclear in HCC. Materials and Methods: Expression of miR-92a in human HCC cell lines was evaluated using qRT-PCR. MTT assay and transwell assay were used to examine the function of miR-92a in HepG2 and ...
متن کاملmiR-26b enhances radiosensitivity of hepatocellular carcinoma cells by targeting EphA2
Objective(s): Although low-dose radiotherapy (RT) that involves low collateral damage is more suitable for hepatocellular carcinoma (HCC) than traditional high-dose RT, but to achieve satisfactory therapeutic effect with low-dose RT, it is necessary to sensitize HCC cells to irradiation. This study was aimed to determine whether radiosensitivity of HCC cells can be enhanced using miR-26b by tar...

متن کاملMicroRNA-424 Is Down-Regulated in Hepatocellular Carcinoma and Suppresses Cell Migration and Invasion through c-Myb
Hepatocellular carcinoma (HCC) is one of the leading causes of cancer-related mortality worldwide. MicroRNAs (miRNAs) are important regulators of multiple cellular processes, and the aberrant miRNAs expressions have been observed in different types of cancer including HCC. Their pathysiologic role and their relevance to tumorigenesis are still largely unknown. In this study, we demonstrated the...
